The leading principle of glandular therapy is similia stimilibus curatur, or "like cures like". Glandular extracts are rich in signalling factors that regulate functions of the corresponding gland, which leads to optimal physiological responses.
The thymus, a small gland in the upper chest, is the key regulator of immunity. It weighs one-third to one-half ounce at birth, and reaches its peak weight of about 17 ounces at puberty. Thereafter, under the influence of many factors, including adrenal and sex hormones, the active thymus gland cells begin to die off, with much of the thymus gland tissue being gradually replaced by fat and connective tissue.
The human thymus produces at least seven thymic proteins; thymopoietin, thymosin alpha, thymulin, thymic humoral factor, hormonal thymic factor, serum thymic factor and other thymic factors. The thymus can enhance, restore, and balance immunity in the following ways:
- Increase the number of lymphokines, including interleukin 2 (IL2), interferon, colony stimulating factor, and others. Thymus gland hormones can cause greater numbers of T cells to develop more IL2 receptors more quickly, which is critically necessary to allow rapid white blood cell proliferation and activation to fight invading germs
- Prevent tissue wasting that occurs with thymus gland removal or severe thymus gland atrophy, and promote healthy weight gain in disease states- such as AIDS- where catabolic body wasting is typical
- Reduce autoimmune reactions
- Prevent the bone marrow injury and subsequent reduction in white and red blood cell production
- Increase disease-fighting antibodies when they're needed, yet reduce the levels of the "allergic antibody" IgE, in patients suffering allergic rhinitis, asthma, and atopic dermatitis
Some of the positive health benefits of the thymus gland hormones may relate to the parallel and synergistic interactions between the pituitary and thymus gland, and growth hormone (GH) and thymus gland hormones. Thymus gland hormones increase the number, activity, and integrity of T cells which secrete GH and GH releasing hormone. Also, they can stimulate the pituitary to release GH.

Thymus extract is non-toxic and side effect free, with few contraindications for use. It should not be used during pregnancy, and should not be used by those with thymic tumours, myasthenia gravis, untreated hypothyroidism and those receiving immunosuppressive therapy- e.g. to prevent transplanted organ rejection.
Liquid thymus extracts are derived from juvenile calves for oral administration. It is important to find a liquid that is concentrated and standardized for peptide content with multiple thymic factors with molecular weights less than 10,000 Daltons.
Some liquid extracts use an exclusive fractionation process that allows the break up of cell membranes and the ability to extract the desired proteins. They are then filtered according to their molecular weight, less than 50,000 Daltons. These procedures are carried out at low temperature near zero degrees C to preserve the integrity of the peptides.
Administration is via the sublingual route allowing for absorption via the mucosae of the mouth. This avoids degradation by the gastrointestinal tract and preserves the properties of the proteins.
